Main Methods

  • Advanced microscopy on embedded tissue sections (cryo-sections & wax-sections) with the confocal laser scanning microscope (CLSM)
  • 3-dimenional fluorescence in situ hybridisation (3D-FISH) for symbiont quantification
  • Post-processing of visual data via image analysis software (daime, PHLIP, imageJ)
  • Quantitative PCR (qPCR) of functional genes (recA) and phylogentic marker genes (16S & 18S) of the symbiont and host
  • Reconstruction of the symbiont phylogeny with molecular tools and sequence analysis with ARB
3D-FISH image of intracellular symbionts
FISH image of the symbionts inside the mussel gill
